4.2 Hvem er kultureliten?
A otimização das variáveis SBSE permitiu o desenvolvimento do método SBSE/LC-UV, o qual apresentou sensibilidade analítica adequada para a análise dos antidepressivos em níveis terapêuticos.
Segundo a validação analítica do método SBSE/LC-UV, realizada em diferentes concentrações plasmáticas, considerando o intervalo terapêutico dos fármacos, o método padronizado, para a análise simultânea dos antidepressivos: mirtazapina, citalopram, paroxetina, imipramina, amitriptilina, fluoxetina, sertralina e clomipramina em amostras de plasma, apresentou seletividade, precisão inter ensaios com coeficientes de variação menores que 15%, recuperação absoluta na faixa de 76% a 100,2%, linearidade de intervalos terapêuticos a tóxicos (de 50 a 1000 ng mL-1) e limite de quantificação adequado. Dessa forma, podemos assegurar que a metodologia padronizada apresenta sensibilidade, repetitividade e exatidão adequadas para fins de monitorização terapêutica de antidepressivos tricíclicos e não- tricíclicos.
O método SBSE/LC-UV também poderá ser empregado na avaliação de níveis plasmáticos, em análises toxicológicas de urgência; após administração acidental de elevadas doses ou em casos de suicídios.
Dessa forma, a técnica SBSE destaca-se como uma alternativa promissora para a substituição das técnicas convencionais. No entanto, o revestimento de polidimetilsiloxano é a única fase extratora disponível no comércio, o que limita a seletividade/especificidade do processo de extração. Assim, é necessário o desenvolvimento de novas fases extratoras (SBSE) mais seletivas e de baixo custo.
